- Work cat: JAK-STAT pathway in disease, 2009:p. 24 (The JAnus Kinase protein and the Signal Transducer and Activator of Transcription represent two core components of the JAK/STAT signal transduction cascade. Originally identified on the basis of the cellular response to interferons, four JAKs (JAK1,2,3 and TYK2) and seven STATs (STAT1,2,3,4,5a,5b and 6) have since been identified in vertebrate systems along with a large and diverse range of trans-membrane receptors and extracellular ligands ... . In addition to the identification of pathway components, considerable research has also produced insights into the roles that the pathway plays during development, immune responses, hematopoiesis and numerous cancers)
- Nature, May 31, 2002:p. 1653 (The Janus kinase-signal transducer and activator of transcription (JAK-STAT) pathway transmits information received from extracellular polypeptide signals, through transmembrane receptors, directly to target gene promoters in the nucleus, providing a mechanism for transcriptional regulation without second messengers)
- Journal of cell science, Mar. 15, 2004:p. 1281 (The JAK/STAT signaling pathway ... The Janus kinase/signal transducers and activators of transcription (JAK/STAT) pathway is one of a handful of pleiotropic cascades used to transduce a multitude of signals for development and homeostasis in animals, from humans to flies. In mammals, the JAK/STAT pathway is the principal signaling mechanism for a wide array of cytokines and growth factors. JAK activation stimulates cell proliferation, differentiation, cell migration and apoptosis. These cellular events are critical to hematopoiesis, immune development, mammary gland development and lactation, adipogenesis, sexually dimorphic growth and other processes. Predictably, mutations that reduce JAK/STAT pathway activity affect these processes ... . Conversely, mutations that constitutively activate or fail to regulate JAK signaling properly cause inflammatory disease, erythrocytosis, gigantism and an array of leukemias)
- Journal of immunology, Mar. 1, 2007:p. 2623 (Universal and essential to cytokine receptor signaling, the JAK-STAT pathway is one of the best understood signal transduction cascades. Almost 40 cytokine receptors signal through combinations of four JAK and seven STAT family members, suggesting commonality across the JAK-STAT signaling system)
- Journal of rheumatology, Sept. 2005:p. 1650 (Jak-STAT signaling occurs when hematopoietins bind to cytokine receptors, initiating a conformational change in the receptor. This brings Jak into apposition, resulting in transphosphorylation and subsequent activation. Once activated, Jak mediate phosphorylation of specific tyrosine residues. STAT and other molecules that recognize these phosphorylated sites are recruited to the receptor and undergo activation by Jak-driven tyrosine phosphorylation. The activated STAT then dissociate, undergo dimerization in the cytoplasm, translocate to the nucleus, and bind to members of GAS ([gamma]-activated site) enhancers)
